Background: Melanotan II is a synthetic analog of the endogenous hormone α-MSH (alpha-melanocyte-stimulating hormone). It was originally developed to induce tanning of the skin without sunlight. Mechanistically, Melanotan II binds to melanocortin receptors in the body – notably MC1R on melanocytes, which stimulates melanin production (melanogenesis) to darken the skin. As a result, users develop a tan complexion as if they had UV exposure, providing a potential protective pigmentation. Interestingly, Melanotan II is non-selective and also activates MC4R (and MC3R) in the brain, which leads to other effects: most prominently increased sexual arousal and libido, and often suppressed appetite. (One of its derivatives, Bremelanotide, was in fact developed and FDA-approved as a treatment for hypoactive sexual desire disorder in women.
