Background: Tirzepatide is a groundbreaking dual-incretin peptide medication that activates both the GLP-1 receptor and GIP receptor. By combining these two hormone pathways, tirzepatide powerfully enhances insulin secretion, suppresses glucagon, and reduces appetite, leading to improved blood sugar control and substantial weight loss. Clinical status: Approved in 2022 for type 2 diabetes (brand name Mounjaro), tirzepatide has also demonstrated remarkable efficacy in obesity trials. In the large SURMOUNT-1 trial, weekly tirzepatide injections produced dose-dependent weight reduction: at the highest dose (15 mg), patients lost about 21% of their body weight over 72 weeks on average (≈20.9% vs ~3% with placebo). Even the 5 mg dose led to ~15% weight loss. These results are unprecedented for a medication, with many participants losing over 50–60 pounds. Importantly, over 85–90% of patients achieved at least 5% weight loss, and more than half achieved ≥20% weight loss at higher doses. Tirzepatide’s dual action is believed to confer greater weight loss than traditional GLP-1 agonists alone. Beyond weight, it significantly improved metabolic health markers: waist circumference, blood pressure, and even delayed progression to diabetes in prediabetic individuals.
